South San Francisco, CA, US
Position Summary:

We seek a highly motivated and experienced protein chemist / biochemist to join the Therapeutic Antibody group at Achaogen to make significant contributions to drug discovery projects targeting disease areas with high unmet medical need. The successful candidate will operate in a highly diverse and dynamic research environment supporting early research and drug discovery programs. The successful candidate will enable drug discovery projects through the expression, purification and characterization of high quality recombinant proteins and antibodies. Attention to detail, the ability to communicate across multiple scientific disciplines and a persistent drive for results are critical to the role.

Key Attributes:

Attention to detail and rigorous observation of variances in methods and controls.
Intense drive to make progress on impactful research programs.
Excellent communicator within and outside of technical area of expertise.

Essential Functions:

Develop and implement creative strategies for high throughput solutions and optimize workflow of existing methods for Achaogen’s protein expression, purification and characterization needs.
Generate protein reagents for immunizations, assay development and target validation.
Express, purify and characterize antibody candidates derived from internal discovery platform at small- and mid-level scale.
Analytical characterization of proteins: endotoxin measurements, electrophoresis, protein quantitation assays, SEC analysis, mass spectroscopy data, antibody affinity measurements.
Independently operates analytical instruments such as spectrophotometers, analytical FPLC (AKTA Pure), plate readers, light scattering systems.
Formulation buffer preparation (molar concentration quantitation and pH adjustment) and optimization.

Essential Experience:

Experience with automation, high-throughput techniques and robotics.
A proven record of success in the expression, purification, and characterization of mammalian proteins, including antibodies, antibody fragments, and novel protein constructs.
Expertise in the use of mammalian and/or insect cell expression platforms, purification systems and analytical instruments (e.g., HEK293 and/or CHO transfections, AKTA/FPLC).
Demonstrated ability to enhance protein production capabilities through identification of key technology improvements, innovation or problem solving.

Preferred Experience:

Antibody binding characterization using Octet or Biacore instrumentation.
Experience in the expression, purification and characterization of bispecific antibody constructs is a plus.
Demonstrated success in a dynamic research environment where adaptability to new projects and approaches was essential.
0-5 years of research experience in an industry or academic setting with an emphasis on protein and antibody production and analysis.

Education:

Minimum of a BS or MS degree in a relevant scientific field (e.g. biochemistry or similar).
